Multi-lab study results of a new recombinant ELISA immunoassay for antibodies to HIV compared against DuPont Western blot.

Haden B, VanWinkle M, Scott M, Busch M, Ascher M, Thorstensson R; International Conference on AIDS.

Int Conf AIDS. 1989 Jun 4-9; 5: 922 (abstract no. E.718).

Beckman Instruments, Inc., 200 S. Kraemer Blvd, Brea, CA 92621, USA

The etiologic agent implicated as the probable cause of Acquired Immunodeficiency Syndrome (AIDS) is the Human Immunodeficiency Virus (HIV). The Beckman ASQ (TM) HIV Reagent Kit is a semi-quantitative assay for the detection & identification of antibodies in human serum or plasma to proteins representing CORE (p24 and Kp 55), ENV (Kp 41, Kp 120N & Kp 120cc) & POL (Kp 66/31). These antigens are purified recombinant peptides produced by genetic engineering techniques in E. Coli. Individual peptides are presented in a microtiter plate format. After incubation of diluted sample (1:100) in the microtiter plate, the plate is washed, developed with alkaline phosphatase conjugate/substrate & read at 410/490 nm. Total assay time is less than 4 hours. The absorbance cutoffs for each peptide were established with 1050 samples. Using the established criteria of positivity for ASQ, 1840 samples were compared against DuPont Western Blot. TABULAR DATA, SEE ABSTRACT VOLUME. Beckman recommends that each evaluator establish their own cutoff & criteria of positivity for this research use only product but found excellent results with the stated criteria in correlation to DuPont Western Blot.
