The Candida albicans activation of anti-U937 cytotoxicity decays progressively with the stage of HIV infection.

Mucosal candidiasis is one of the first infections during the HIV disease. A mannoprotein fraction (MP-F2) from Candida albicans was as efficient as IL-2 in generating cytotoxicity against uninfected orHIV-persistently infected U937 cell line in cultured lymphomonocytes (PBMC) from normal donors. MP-F2-activated killing (MAK) of U937 decreased progressively with the stage of HIV infection, together with the production of TNF-alfa, GM-CSF and IFN-gamma by PBMC from HIV positive subjects. That the decreased production of IFN-gamma is crucial to the decay of U937-MAK generation is suggested by the observation that the addition of anti-IFN gamma antibodies abolished both U937-MAK generation in PBMC from healthy subjects, IFN-gamma activity and TNF-alfa production. The addition of recombinant IFN-gamma markedly increased U937-MAK generation in PBMC of stage II and III subjects.
