Evaluation of peptide-based HIV ELISA kits.

OBJECTIVE: The aim of this study is to verify whether or not NB-02(for detection of HIV-1 and/or-2 antibodies) and NB-06 (for differentiation between HIV-1 and -2 antibodies) kits are useful as screening and differentiation tests using an exclusive automatic analyzer. METHODS: Two kinds of kit(mentioned above) and the exclusive automatic analyzer were obtained from BIOCHEM IMMUNOSYSTEMS INC. (Canada). The principle of these kits is ELISA for detection of IgG-Ab. Experimental items: 1)Evaluation of NB-02 for its specificity, reproducibility and sensitivity; 2) Etiological study on HIV-1 and/or HIV-2 infections in sera of several races in combination with NB-02 and NB-06. RESULTS: (1) Evaluation of NB-02; Specificity: Anti HIV-positive samples tested (n = 75) were seropositive by this kit, and the seropositivity was markedly correlated with that by the Serodia-HIV kits (PA method) which are widely used as a screening test in Japan; Reproducibility: C.V. of the 3 lots were 3.4, 6.0, 3.2%, respectively; Sensitivity: HIV-positive samples (n = 75) prediluted 10(4)-fold exhibited to be highly reactive with the percentage of 96%. (2) It was found that Zambian (n = 28), Thai(n = 11) and Japanese(n = 19) sera tested were all infected with HIV-1 alone, whereas Senegalese sera tested (n = 11) were mostly infected with HIV-2, except for one sample (co-infection with HIV-1 and -2). DISCUSSION AND CONCLUSIONS: As to the performance of NB-02 and NB-06 kits and the exclusive automatic analyzer: 1) The operation with autosystem was simple; 2) The determination was rapid and accurate; 3) The sensitivity and the reproducibility were satisfactory. From these results NB-02 and NB-06 using the exclusive automatic analyzer are useful for routine assay in hospitals and clinical laboratories.
