The activity of vaccine adjuvants can generally be attributed to their physical or chemical properties, which alter the route or rate of antigen delivery to the immune system or directly activate lymphoid cells. Nonionic block copolymers, synthesized from repeating units of oxypropylene and oxyethylene, can be designed so that individual copolymers have unique physical properties with differential levels of adjuvant activity. We have designed block copolymers that can assemble into 1-2 micrometer particles when formulated with protein antigens in aqueous solutions. These copolymers are potent adjuvants which appear to (1) alter the route of antigen delivery and (2) directly activate cells. An individual synthetic copolymer can be used to augment both antibody and cellular immune responses, including the induction of cytotoxic T-lymphocyte responses. Because of their compatibility with aqueous buffers, these copolymers are nontoxic and can be formulated with a wide variety of protein antigens.
