OBJECTIVES: To compare frequency of T cells with an antigen-presenting-cell phenotype in peripheral blood of HIV patients and healthy seronegative individuals. METHODS: 20 adult HIV patients and 10 HIV-negative controls were included in this study. We measured expression of CD80, CD86, CD28 and HLA-DR molecules on CD4+ and CD8+ T cell subsets in peripheral blood by means of three-colour flow cytometry. RESULTS: A low CD80 and CD86 expression (mean CD3/CD80: 4.97%, mean CD3/CD86: 0.8%) was found in HIV-negative controls. In HIV-patients CD80 and CD86 molecules were present on both T cell subsets (mean CD4/CD80: 5.0% and mean CD4/CD86: 2.6%, mean CD8/CD80: 4.1% and mean CD8/CD86: 2.7%) and were associated with HLA-DR expression. Only the expression of CD86 was significantly (p < 0.05) increased on T cells of HIV-infected individuals. The expression of CD28 was decreased on T cells of HIV patients (mean CD28: controls 89.8%, HIV-infected 49.7%) and was negatively correlated to the expression of HLA-DR and CD86 (correlation coefficient: -0.75, -0.71). CONCLUSION: The increased expression of costimulatory molecules CD80 and CD86 on T cells in peripheral blood of HIV patients compared to HIV-negative controls suggests that T cells develop an antigen-presenting-cell phenotype in HIV-infection, which might contribute to immune mediated destruction of CD4+ T cells.
