BACKGROUND: Toll-like receptors (TLR) are known to induce innate immune responses upon stimulation by a wide variety of pathogens. It has recently been shown, that TLR4 plays a role in host defense against respiratory syncytial virus (RSV) infection. Aim and METHODS: To determine the role of TLR4 in the innate immune response to infection with the RNA viruses influenza (A/PR/8/34) and parainfluenza (Sendai-strain), C3H/HeJ (TLR4 mutant) and C3H/HeN (wt) mice were intranasally inoculated with either 10 TCID50 influenza or 10e6 TCID50 parainfluenza. On day 1, 4, 8 and 14 postinfection, viral load was determined by real-time PCR, cytokines by ELISA, and cell-infiltrates by cytospin. Statistics by Mann-Whitney. RESULTS: Infection with either influenza or parainfluenza resulted in an initial increase in viral loads in lungs peaking on day 4, and a rapid viral clearance thereafter. Influenza and parainfluenza viral loads were similar in TLR4 mutant and wt mice at all time points. In addition, recruitment of leukocytes to the lung and increased production of IL-6, IL-12 and IFN-g occurred to a similar extent in TLR4 mutant and wt mice in both infection models. Conclusion: Although RSV, parainfluenza (paramyxoviridae) and influenza (orthomyxoviridae) belong to the same superfamily of RNA viruses, TLR4 does not play a role in the innate immune response to respiratory tract infection with influenza or parainfluenza virus.
