Background: Malignant pleural mesothelioma (MPM) is a rare and rapidly progressive malignancy of the mesothelium involving the pleura (lining of the lungs). Currently in the UK there is no accepted standard treatment regimen for MPM. Pemetrexed disodium in combination with cisplatin (administered with vitamin B12 and folic acid) is the first and only chemotherapy agent licensed in the UK for use for MPM treatment. Objectives: To assess the clinical effectiveness of pemetrexed disodium in combination with cisplatin for the treatment of unresectable MPM in chemotherapy naive patients. Methods: The review was conducted following accepted guidelines for conducting systematic reviews including the identification studies (1980 to may 2005), application of inclusion criteria, quality assessment of included studies and data extraction and analysis. Results: One randomised controlled trial (RCT) comparing pemetrexed plus cisplatin with cisplatin alone, involving a total study population of 448 patients, fulfilled the inclusion criteria. No RCTs comparing the effectiveness of pemetrexed with other commonly used alternatives such as vinorelbine, MVP (mitomycin C, vinblastine and cisplatin) or supportive care were identified. The presentation will summarise key results including treatment-related adverse events (as relative risks with confidence intervals); data availability and study quality and implications for policy makers.
